    摘要:

    miRNA-21 在大部分肿瘤中都存在高表达,其含量的异常可作为 肿瘤发生的早期诊断标志。基于 G- 四链体 -hemin DNAzyme 能催化 H 2 O 2 氧化 2, 2’- 联氨双(3- 乙基苯并噻唑啉 -6- 磺酸)二铵盐(ABTS)生成 ABTS + ,利用链的竞争作用构建了 G4-DNAzyme 比色生物传感器,该传感 器检测 miRNA-21 时具有操作简单、响应快速的特点。在优化核酸序列设 计、反应温度、反应时间等实验条件的基础上,利用紫外分光光度计测量 ABTS + 的最大吸光度,并计算其与空白样品的信背比(SBR),在不同浓度 的 miRNA-21 下,绘制浓度 -SBR 曲线。结果显示:实验可检测 miRNA-21 的最低浓度约为 0.1 μmol/L;在 miRNA-21 浓度为 0.1~1.2 μmol/L 时,SBR 与 miRNA-21 的浓度呈现出良好的线性关系,其线性方程为:y=0.834 9x+ 0.898 3,相关系数 R 2 =0.995。该 G4-DNAzyme 比色生物传感器设计简单, 操作方便,无需高精密仪器,对目标物有良好响应性及高选择性。

    Abstract:

    MiRNA-21 is highly expressed in almost all tumors, and abnormal miRNA-21 content can be used as an early diagnostic marker for tumor. G-quadruplex-hemin DNAzyme can catalyze the oxidation of 2, 2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)to ABTS + by H 2 O 2 , and based on chain replacement, a visual detection sensor with simple operation and fast response for miRNA-21 was constructed. After optimizing nucleic acid sequence, reaction temperature and time, measuring maximum UV absorbance of ABTS + with different concentrations of miRNA-21 and calculating the SBR with the blank sample, the concentration-SBR curve was drawn. It was found that the lowest concentration to detect was 0.1 μmol/L, when the concentration of miRNA-21 was within the range from 0.1 to 1.2 μmol/L, there was a good linear relationship between SBR and miRNA-21 concentration with the linear equation being y=0.834 9x+ 0.898 3,the linear correlation coefficient being R 2 =0.995. Meanwhile, this colorimetric biosensor was simple in design and operated with high selectivity for detecting miRNA-21.
