Abstract:By high throughput detection means, the single nucleotide polymorphisms (SNPS) analysis is conducted on metallothionein MT-2A gene promoter core region of metallurgy operation population. The MT-2A gene promoter sites of metallurgy workers (288 cases) are carried on high throughput SNP detection by magnetic nanoparticles microarray technology, and the sequencing test is also made. The results show that three genotypes (AA, AG and GG) frequency of the MT-2A gene promoter core region in the workers are 87.4%, 11.8% and 0.8%, and alleles (A and G) frequency are 93.2% and 6.8%, respectively. The MT-2A genotype and allele frequencies are consistent with Hardy-Weinberg equilibrium (p>0.05). Finally applies the direct sequencing method for PCR product sequencing test, and the obtained results are consistent with the result of SNP analysis.
